  • 激素或細胞因子可以透過活化受體的多樣化訊息來影響不同的細胞,進而建構出各種的生理反應,因此,也是生醫藥物開發的重點。目前有超過90%的市售藥物都是針對調控它們的訊息機轉而設計出來的衍生物。本實驗室致力於以分子生物學的技術來探討激素和細胞因子訊息的相關機轉。目前主要目標放在蛋白質性質的分子,以期了解它們如何透過受體的整合訊息去影響生理反應或疾病 (如癌症等) 的相關進程。另外,透過基因體學分析或遺傳工程重組技術,我們也有興趣於尋找或開發新型的蛋白質激素和細胞因子,以評估其生物活性和未來是否有機會作為蛋白質候選藥物。以下簡要說明實驗室的幾個研究主題:

    Project 1. TGF-beta/BMP 訊息對於幹細胞分化的影響。

    TGF-beta/BMP家族的成員對於幹細胞的幹原性、細胞分裂、分化和凋亡等各種能力有著不同層面的影響。本實驗室在這個主題上的研究方向略分為二:其一是針對它們如何主導間質幹細胞 (mesenchymal stem cells) 的成骨分化和脂肪分化能力;其二是針對它們對於生殖幹細胞與體細胞的功能影響。我們希望全面性地了解這個家族成員在骨頭或生殖系統的微環境中的表現消長,進而解析個別分子的訊息和功能,希望未來能在骨癒合或不孕症等領域有其應用價值。

    Project 2. 卵巢生理調控與生殖系統相關癌症的致病機轉

    根據分析,哺乳動物有超過70%以上的基因都會在卵巢週期的特定期間中有所表現,因此卵巢一直是研究基因體學的寶厙。我們的研究策略是利用基因體分析,找出在卵巢週期中具有明顯變化的蛋白激素或受體基因,進而以分子層次研究其訊息對於週期、卵子品質、不孕症等的影響。另外,卵巢癌為婦科癌症中致死率最高且復發率最高的癌種,且具有獨特的腹水轉移機轉。本實驗室也致力於以細胞訊息層次,搭配實驗動物模型來研究卵巢癌,藉此希望更加了解其致病機轉,並對卵巢癌或其他生殖系統癌症的診斷和治療有所幫助。

    Project 3. 以基因體分析或基因重組技術開發活性蛋白分子或候選藥物

    蛋白質通常具有保留的模版以形成特殊構型和功能區塊。本實驗室藉由找出蛋白質激素或因子所屬的功能模版,即可在不同的基因體資料庫中搜尋出相關的未知蛋白;或將不同模版重組,改變激素或因子的生物特性,藉此開發出新型蛋白分子,以分析它們的生物功能或評估作為蛋白質候選藥物的可能性。相關策略如下圖。
