  • 間質幹細胞分化機轉、生殖系統生理與癌症、激素或細胞因子藥物開發、訊息傳遞等分子內分泌學
  • 研究方向
  • 激素或細胞因子是主導各種生理反應的重要分子,一直以來都是生醫研究和藥物開發的重點標的。本實驗室的研究重點有二:(1)致力於以分子內分泌學的技術來探討蛋白質類的激素和細胞因子如何透過對應受體的整合訊息去影響生理反應、疾病 (如癌症等) 或幹細胞分化的相關進程;(2) 透過基因體學分析和遺傳工程重組技術,開發或設計新型的蛋白質激素和細胞因子,以評估其機轉、生物活性和成為蛋白質候選藥物的潛力。以下簡要說明幾個研究主題:

    Project 1. TGF-beta/BMP 訊息對於幹細胞分化與細胞功能的影響。

    本實驗室利用解析細胞訊息、佐以原代細胞培養、小鼠生理模式、疾病相關體學分析等,探討TGF-beta/BMP家族的訊息對間質幹細胞 (mesenchymal stem cell)、生殖幹細胞與體細胞的功能影響。希望未來能在骨癒合、骨質疏鬆或不孕症等領域有其應用價值。

    Project 2. 卵巢生理調控與生殖系統相關癌症的致病機轉

    卵巢除了是掌控生理和生殖的重要器官之外,其衍生癌症也是婦科癌症中致死率最高且復發率最高的癌種。我們的研究策略是利用基因體分析,找出在卵巢週期或癌症病程中具有明顯變化的蛋白激素或受體基因,進而設計細胞模式、原代細胞實驗以操控細胞訊息與基因表現、再佐以小鼠活體驗証等,藉此了解特定的基因功能和卵巢癌致病機轉。

    Project 3. 開發新型的蛋白質激素或候選藥物

    蛋白質型藥物具有比一般化學藥物更高的受體結合專一度,因此具有極佳的生理相容性和極低副作用。本實驗室的策略是利用重組和突變等技術改良具有蛋白質型藥物潛力的激素或細胞因子,試圖克服其折疊效率低和生理半衰期短的缺點,並佐以細胞訊息分析和小鼠實驗評估其作為候選藥物的潛力。目前的研究重點在胱胺酸結狀蛋白 (cystine-knot protein) 系列,例如長效型血管內皮細胞生長因子已進入專利審查階段。
